SOP for Microscope

1. Purpose
To lay down the procedure for proper handling, operation, cleaning, and maintenance of microscopes to ensure accurate and reliable observations.
2. Scope
This SOP is applicable to all microscopes (compound, stereo, digital, or phase-contrast) used in the laboratory/quality control/microbiology department.
3. Responsibility
-
User/Analyst: Proper usage, cleaning, and reporting any malfunction.

Lab Supervisor/Manager: Ensuring adherence to SOP, training, and arranging maintenance.

Engineering/Maintenance Team: Preventive and corrective maintenance.
4. Materials & Accessories
-
Lint-free cloth or lens paper
-
Lens cleaning solution (or 70% IPA for cleaning surfaces, not lenses)
-
Dust cover
-
Logbook for usage/maintenance record
5. Procedure
5.1 Pre-Operation Checks
-
Ensure microscope is placed on a clean, vibration-free, dry surface.

Check power supply (for electrical microscopes).
-
Verify cleanliness of eyepieces, objective lenses, and stage.
-
Ensure dust cover is removed and stored properly.
5.2 Operation
-
Switch on the microscope (for electric light source models).
-
Place the prepared slide on the stage and secure with clips.
-
Start with the lowest magnification objective (usually 4X or 10X).
-
Adjust coarse focus knob slowly until the object is visible.
-
Use the fine focus knob for sharp clarity.
-
Adjust diaphragm and light intensity for proper illumination.

Shift to higher magnifications (40X, 100X oil immersion) as needed:

For 100X oil immersion: Add a drop of immersion oil and carefully focus.

Record observations in laboratory notebook or system.
5.3 Post-Operation
-
Remove the slide carefully and clean stage if any spillage occurs.

Wipe off oil from 100X objective lens with lens paper immediately after use.

Switch off light source and unplug microscope.
-
Cover microscope with dust cover.
5.4 Cleaning & Maintenance
-
Clean lenses only with lens paper and suitable lens solution.
-
Do not use tissues, cloth, or fingers.
-
Weekly: Wipe body with dry lint-free cloth.
-
Monthly: Inspect for loose screws, light alignment, and smooth knob movement.

Annually: Calibrate microscope and perform preventive maintenance by authorized service engineer.
6. Precautions
-
Never touch lenses with bare fingers.
-
Do not expose microscope to direct sunlight, moisture, or corrosive chemicals.
-
Always start focusing with low power objective.
-
Avoid dragging microscope; lift with both hands (arm & base).
7. Documentation
-
Record microscope usage in logbook (Date, User, Purpose).
-
Document cleaning and maintenance activities.
-
Report and record any malfunction immediately.
