Standard Operating Procedure (SOP) for Disposal of Empty Media Containers
1.0 Purpose
To establish a standard procedure for the safe and compliant disposal of empty media containers used in microbiology laboratories, in order to prevent contamination and ensure environmental safety.
2.0 Scope
This SOP applies to all laboratory staff involved in the handling and disposal of empty culture media containers (bottles, tubes, flasks, and plates) in the QC Microbiology Laboratory.
3.0 Responsibility

Analyst/Operator: To ensure proper handling and disposal of empty media containers as per this SOP.

Microbiology/QA Head: To review records and ensure compliance.

Housekeeping/Support Staff: To assist in final disposal after decontamination.
4.0 Definitions
-
Media Containers: Glass or plastic bottles, tubes, Petri plates, or other containers previously used for preparation or dispensing of culture media.

Decontamination: Process of rendering containers free from viable microorganisms (autoclaving/disinfecting).
5.0 Procedure
5.1 Collection
-
Collect empty media containers immediately after use.
-
Ensure containers are free from unused or leftover media. If media residue is present, autoclave before disposal.

Place empty containers in a designated, labeled tray or bin marked “Used/Empty Media Containers – To Be Disposed”.
5.2 Decontamination
-
For glass containers:
-
Rinse with sufficient water to remove media traces.

Autoclave at 121°C, 15 psi for 30 minutes before final disposal/reuse.

For plastic containers (single-use):

Ensure all residues are removed.

Autoclave or treat with suitable disinfectant (e.g., freshly prepared 1% sodium hypochlorite) for at least 30 minutes.

After treatment, discard as per biomedical waste guidelines.

5.3 Disposal

Glassware:
-
After autoclaving, segregate reusable glassware for washing, drying, and sterilization.

Broken glassware must be discarded in a puncture-proof sharps container after autoclaving/disinfection.

Plastic containers:

Dispose of in yellow biomedical waste bags (as per local regulatory norms) after disinfection/autoclaving.

5.4 Documentation
-
Record details of autoclaving/disposal in the Autoclave Logbook or Waste Disposal Logbook.

Maintain traceability with date, analyst name, and quantity of disposed containers.
6.0 Precautions
-
Always wear PPE (gloves, lab coat, goggles) while handling used containers.

Never dispose of containers without prior decontamination.
-
Avoid direct hand contact with potentially contaminated surfaces.
-
Ensure segregation of reusable and non-reusable containers.
